This might have to do with your Silverlight permissions settings -- open Silverlight and check the permissions tab to see if you or a firewall blocked the app somehow.

Yes, SketchUp user-interfaces on Windows run inside of Internet Explorer, so check out your settings in Control Panel > Internet Options and make sure the Microsoft Silverlight plugin is not disabled or disallowed.

Go (in Windows, not SketchUp) to Start > Control Panel > Internet Options. Most browser restrictions are administered from there, and you can find lots of answers via Google for this error (e.g. here) which should help you check out your configuration. Keep in mind that this may not be something that you did -- 3rd-party antivirus/anti-malware programs may change your browser security settings too.
