Pre-order V5 from V3 or V2

If we pre-order an upgrade from Version 2 or 3, do we have access to Version 4 until Version 5 is officially released?

No, we don't deliver Maxwell 4 license until Maxwell 5 is realeased. But it's not that far. I mean, we have a sign in the wall of days until release. Days, not months. But for those concerned about pre-order ending before they decided, we will say when it's going to finish before hand.

after update from v4 to v5 will be possible to use v4? or only v5?

Asked the same question to support myself when I pre-ordered version 5. Support told me "yes" I'd be able to continue using version 4, no problem at all. Hope that helps you out.
I'm excited about the release of version 5 myself. Looking forward to all the new features on the horizon. It looks well worth the wait and the pre-order discount is pretty nice too. (Note that you'll need to upgrade the render nodes package as well if you want it --those are now a separate thing.)
